INFLUENCE OF USE N-ACETYLCYSTEIN FOR THE CORRECTION OF ANTIOXIDANT STATUS IN RABBITS’ ON INDICATORS OF BLOOD PLASMA LIPID PROFILE

Introduction. The lipid profile of the blood plasma is an important parameter that must be determined during medical examination. Abnormal plasma lipid levels reflect the onset or worsening of problems associated with metabolic diseases such as obesity, liver lipidosis, diabetes mellitus, and atherosclerosis. Described processes are accompanied by the development of oxidative stress, which can be corrected with the use of antioxidant compounds, for example, N-acetylcysteine, an amino acid with pronounced redox properties as an antioxidant protector against cadmium load, arsenic poisoning and other toxicants, which has a positive effect on reproductive capacity.

The goal of the work was to establish the peculiarities of the lipid profile in the blood plasma of intact rabbits and to correct their antioxidant status with N-acetylcysteine.

Materials and methods. The studies were performed on sexually mature male rabbits of the Hyla breed (n=14). The groups were formed according to the principle of analogues, 7 animals each. The experimental group was orally administered N-acetylcysteine at a dose of 50 mg/kg for 21 days once. Males of the control group were injected 0.9% sodium chloride solution according to the same protocol.

Blood was taken from the ear artery of each rabbit 21 days after the start of the experiment. All samples were collected in centrifuge tubes containing anticoagulant and centrifuged at 3000 g for 15 minutes at 4°C to collect plasma. The levels of total cholesterol, triacylglyceroles, high-density and low-density lipoproteins were determined colorimetrically, and total antioxidant activity was determined spectrophotometrically.

Research results and their discussion. The effectiveness of the use of N-acetylcysteine for the correction of antioxidant status in rabbits was evaluated by a complex indicator – the total antioxidant activity of the blood plasma, which in the experimental group was significantly higher than the control data. Thus, their antioxidant status increased, which should have a positive effect on lipid metabolism. Introduction of N-acetylcysteine showed decrease in the level of total cholesterol in the rabbits of the experimental group, as did the amount of triacylglyceroles in the plasma compared to the control values. The improvement of these indicators had an impact on the level of lipoproteins of different densities.

Conclusions and prospects for further research. The use of N-acetylcysteine to correct the antioxidant status in the body of male rabbits has a positive effect on the lipid profile of the blood plasma.

The level of total cholesterol in the rabbits of the experimental group decreased by 9.3% (p≤0.05) compared to the control values, while the amount of triacylglyceroles in the plasma was significantly lower by 13.9% (p≤0.01). The levels of lipoproteins underwent significant changes – amount of low-density lipoproteins was lower by 38.6% (p≤0.001) of the data of the control group, while the level of high-density lipoproteins in the rabbits of the experimental group was reduced by 19.6% (p≤ 0.001).

The effectiveness of the antioxidant effect of N-acetylcysteine is proven by positive changes in the dynamics of the total antioxidant activity of the blood plasma of rabbits – after the end of the experiment, this indicator was 17.8% (p≤0.05) higher than the values of the control group. The perspective of further research is the assessment of the impact of antioxidant correction with
N-acetylcysteine on the quality and fertilizing ability of rabbit sperm, its redox potential.
