COMPARATIVE EFFICIENCY OF CHORIOPTES BOVIS MITES MICROPREPARATIONS PREPARING
Introduction. Acariform parasitic mites belonging to the genus Chorioptes (Acariformes: Psoroptidae) are distributed worldwide, causing chorioptosis in many domestic and wild animals. The diagnosis of chorioptosis in ruminants is based on clinical signs and microscopic detection of mites in scrapings from the skin of the affected animal with mandatory species identification of choriopteses. Genetic diagnosis of choriopteses is expensive and requires quite a lot of time to carry it out, so it is necessary to search for new effective methods of diagnosing chorioptosis.
The goal of the work was to establish the effectiveness of the methods of preparing micropreparations from Chorioptes bovis in a comparative aspect.
Materials and methods. At the first stage of research, the most optimal values of the ratio proposed by us were determined: glycerin and alcohol solutions of iodine (1:1, 2:1, 3:1), which were used as a dye for the production of micropreparations from C. bovis.
At the second stage of research, the effectiveness of the methods of micropreparations preparing were compared, namely: it was proposed the one using an iodine-glycerol mixture and the well-known one using1% alcoholic solution of diamond green as a dye.
Results of research and discussion. The conducted studies established that the most effective method of preparing micropreparations from Chorioptes bovis in terms of the degree of staining of their morphological structures is the method using a mixture of glycerol and an alcohol solution of iodine (in a ratio of 3:1) compared to the method where a 1% alcohol solution of diamond green is used as a dye.
Conclusions and prospects for further research:
1. The most effective method of preparing micropreparations from Chorioptes bovis is the method using a mixture of glycerin and an alcohol solution of iodine in a ratio of 3:1.
2. When comparing the degree of staining of the morphological structures of choriopteses, the method using the iodine-glycerol mixture showed a high degree of staining according to 7 indicators, and the method using a 1% alcohol solution of diamond green – only according to 3 indicators.
The results obtained by us regarding the effectiveness of the methods of preparing micropreparations from choriopteses will be used in the following tests to establish the identification features of C. bovis.
