ANTIVIRAL ACTIVITY AND INTERFERON PRODUCTIVITY OF LOW-MOLECULAR INDUCERS OF INTERFERON ANALOGS OF AMIXIN AND DERIVATIVE OF DIPHENYL 

   Introduction. In Ukraine, the issue of prevention and treatment of mass viral diseases of adult and young animals, which are caused by secondary immunodeficiencies of various origins that weaken the immune status of the body, is acute.

   The study of the biological activity of new interferon inducers and the introduction of the developed drugs and methods of application will allow their use as etiotropic methods of emergency prevention and treatment of viral diseases in productive and non-productive animals, correction of immunodeficiency states of various origins, strengthening of the protective properties of vaccines when vaccinating young animals.

   The goal of the work. Determination of antiviral and interferonogenic activity of low-molecular-weight interferon inducers – Аmiksin analogues and diphenyl derivatives, by studying their biological properties in vitro and in vivo and relating the detected effects to the planarity of their structure.

   Materials and methods. The investigated substances were 11 newly synthesized compounds – diphenyl derivatives, analogs of Amiksin. A low molecular weight interferon inducer, known in medical practice Amiksin, was used as a comparison drug.

   Manipulations with experimental animals (sampling of blood, obtaining macrophages of peritoneal exudate and spleen cells of mice) were carried out in accordance with generally accepted guidelines and compliance with bioethical norms. In order to evaluate the prospects of further research, the toxicity of the drugs was determined in vitro and in vivo. To interpret the results, studies of interferonogenic and antiviral activity in vitro and in vivo were conducted according to generally acceptedguidelines.

   Results of research and discussion. Revealed that in vitro, in L929 cell culture the studied compounds have interferon productivity abilities in doses no higher than effective doses of Amixin (4-31 μg/ml). Demonstrate that the antiviral activity of the investigated compounds caused by their interferon-inducers action in vitro to determine the relationship between the planar structure and biological effects of newly-synthesized compounds.

   We detected the studied compounds (№ 9, № 10 and loramixin) in vivo increases the pool of interferon response of cells more intensely than officinal product – Amixin.New interferon inducers have demonstrated the ability to increase cytokine levels in animals infected with herpes simplex virus. The using of these compounds showed that compounds № 9, № 10 and loramixin are more active antiviral agents compared with аmixin in experimental herpetic infection: the magnitude of the multiplicity of security compound № 9 reaches 100,0; loramixin do not differ from that of the comparison drug (10.0).

   Conclusions and prospects for further research. Toxicity of experimental drugs and their interferonogenic properties are related to the planarity of the substance structure.Investigated polycyclic compounds have several advantages over officinal product Amixin that allows us to consider them as verypromising inducers of interferon and antiviral drugs.
